ABSTRACT

The cardiovascular patch, served as artificial graft materials to replace heart or vascular tissue defect, is still playing a key role in cardiovascular surgeries. The defects of traditional cardiovascular patch materials may determine its unsatisfactory long-term effect or fatal complications after surgery. Recent studies on many new materials (such as tissue engineered materials, three-dimensional printed materials, etc) are being developed. Patch materials have been widely used in clinical procedures of cardiovascular surgeries such as angioplasty, cardiac atrioventricular wall or atrioventricular septum repair, and valve replacement. The clinical demand for better cardiovascular patch materials is still urgent. However, the cardiovascular patch materials need to adapt to normal coagulation mechanism and durability, promote short-term endothelialization after surgery, and inhibit long-term postoperative intimal hyperplasia, its research and development process is relatively complicated. Understanding the characteristics of various cardiovascular patch materials and their application in cardiovascular surgeries is important for the selection of new clinical surgical materials and the development of cardiovascular patch materials.

ABSTRACT

Objective:To probe into Rab25 Gene’s Effect on TGF-β inhibition of proliferation, invasion and epithelial mesenchymal transformation (EMT) of breast cancer MDA-MB-231 cells and explore its molecular mechanism.Methods:The experiment was divided into three groups: control group,TGF-β Group and si-Rab25 group. TGF-β induced MDA-MB-231 cell model of EMT was built. CCK-8 assay was used to detect cell proliferation. Transwell assay was used to detect the ability of cell invasion and migration.Western blot was used to detect the changes of related proteins in each group.Results:After stimulating MDA-MB-231 cells with TGF-β, Rab25 gene was highly expressed. Compared with TGF-β group (57.48±%3.62%), the migration ability and invasion ability of cells in si-Rab25 group (33.49%±2.93%) decreased by 41.7%, with a significant difference ( P<0.05). Compared with TGF-β group (153.21%±4.17%), the proliferation ability of cells in si-Rab25 group (115.32%±5.69%) decreased by 24.73%, with a significant difference ( P<0.05). The expression of MDA-MB-231 fine EMT related protein in si-Rab25 group was significantly different from that in TGF-β group ( P<0.05). The expression of p-AKT and Snail protein in si-Rab25 group was significantly lower than that in TGF-β group ( P<0.05) . Conclusions:Rab25 gene is highly expressed in MDA-MB-231 cells. Silencing Rab25 gene can activate AKT signal pathway, inhibit Snail protein expression, regulate EMT related protein expression, and inhibit EMT transformation.

ABSTRACT

Objective To investigate the status and significance of KIT,PDGFRA and DOG1 gene mutation in gastrointestinal stromal tumors (GIST).Methods 100 GIST patients treated in the First Affiliated Hospital of Guangxi Medical University between May 2002 and May 2013 were analyzed restrospectively.DNA was isolated and amplified for the all exons of KIT,PDGFRA and DOG1.Each PCR product was sequenced to find the position and type of mutation.Results KIT mutations were identified in 75 cases (75%).PDGFRA mutations were found in 16 cases (16%).No DOG1 mutations were found.The overall 5-year survival was 58.8%.Log-rank univariate survival analysis showed that the primary location,tumor size,metastasis,operational mode,type of tumor cells,invasion of the smrounding organs,invasion of the smooth muscle,mitotic counts of the tumor cells,deletions in exon 11 KIT and targeting therapy were significant prognostic factors (all P < 0.05).COX regression model showed that tumor size,metastasis,operational method,invasion of the surrounding organs,mitotic counts of the tumor cehs,deletions in exon 11 KIT and targeting therapy were related to prognosis.Conclusion KIT and PDGFRA mutations are mutually exclusive.The overexpression mechanism of DOG1 is not related to DOG1 gene mutation.The related gene mutations affect the prognosis of GIST.

ABSTRACT

Objective To investigate the the clinical effect and immune regulatory mechanism of red-blue light combined with Niuhuang Shangqing capsules for the moderate-severe acne.Methods A total of 180 patients with moderate to severe acne treated in People's Hospital of Nanhai District in Foshan City from June 2014 to June 2016 were randomly divided into 2 groups with each group 90 patients. The control group was treated with red and blue light, and observation group was treated with the combination of red and blue light andNiuhuang-Shangqingcapsules. The levels of SP(Substance P), IL-1 and IL-6 in peripheral blood were observed and analyzed. The recurrence rates and adverse events were observed.Results After treatment, the SP (657.4 ± 36.6 pg/mlvs.799.9 ± 60.2 pg/ml,t=19.188), IL-1(61.8 ± 24.7pg/L vs.92.1 ± 23.5 pg/L,t=8.431), IL-6 (38.7 ± 10.3pg/mlvs.66.7 ± 14.1pg/ml, t=12.421) of the observation group were significantly better than those of the control group (allPs<0.01). The total effect rate of the observation group were 91.11% (82/90), which were significantly higher than 70.00% (63/90) of control group (χ2=12.804,P<0.01). Conclusions The Red-blue light combined withNiuhuang-Shangqing capsules for treating moderate-severe acne, has significant effect and can reduce the serum levels of SP, IL-1 and IL-6, with fewer adverse events and lower recurrence rates. They can be used as a safe and effective treatment of acne.

ABSTRACT

Objective To study the expression and clinical significance of Mybl2 in Cervical tissues.Methods 74 cases of cervical tissues were obtained from the department of obstetrics and gynecology.The expression level of Mybl2 was detected by Elisa method,and the relationship with clinical pathology was analyzed.Results Elisa results showed that the expression of Mybl2 was significant among inflammation,CINⅠ,CINⅡ,CINⅢ and ICC groups (F=27.39,P<0.05).The expression level of Mybl2 was higher in the older group that the younger one which were in the same clinical pathology degree,but it was not was significant(P>0.05).Conclusion The expression levels of Mybl2 were related with the pathological changes of cervix.Disturb expression level of Mybl2 maybe induce the development of cervical cancer.

ABSTRACT

Objective To compare the distribution and drug resistance of isolates between sterile body fluid and non‐sterile body fluid in the hospital in 2014 .Methods By adopting the retrospective analysis method ,we used BD phoenixTM 100 to conduct bacteria identification and drug susceptibility testing ,the Whonet5 .6 software and SPSS19 .0 software to statistically analysize the drug re‐sistance of the bacteria .Results E .coli ranked the top in sterile body fluid isolates(43% ) while the highest rate in non‐sterile body fluid was P .aeruginosa .E .coli(21% ) .Isolates from sterile body fluid had lower drug resistance rate to 11 kinds of antibacterials such as ampicillin ,chloramphenicol ,ciprofloxacin and aztreonam than the strains isolated from non‐sterile body fluid(P<0 .05) .S . aureus ,isolated from sterile body fluid ,had lower drug resistance rate to 6 kinds of antibacterials such as amikacin ,amoxicillin/cla‐vulanicacid ,ciprofloxacin than the strains isolated from non‐sterile body fluid .P .aeruginosa ,isolated from sterile body fluid ,had lower drug resistance rate to aztreonam than the srains isolated from non‐sterile body fluid .K .pneumoniae ,isolated from sterile body fluid ,had lower drug resistance rate to 6 kinds of antibacterials such as ampicillin/sulbactam ,sulfamethoxazole ,chlorampheni‐col than the strains isolated from non‐sterile body fluid(P<0 .05) .Conclusion There is significant difference between sterile body fluid and sterile body fluid in strain distribution and drug resistance ,so it is vital to enhance the bacterial resistance surveillance of sterile body fluid .

ABSTRACT

Objective:To evaluate influence of electroacupuncture combined point massage on aptients with cartid ar- tery arteriosclerosiss.Methods:A total of 106 patients with carotid atherosclerosis were randomly divided into rou- tine group (n=54,received health education based on routine therapy)and electroacupuncture+ massage group (n=52,received additional electroacupuncture and point massage based on routine group).Both groups were treated for two courses,20 times/course, 1 time/d.Carotid intima-media thickness (IMT)and thickness of carotid plaques were measured and compared between two groups before and after treatment.Results:There were no sig- nificant difference in thickness of carotid plaques and IMT between two groups before treatment,P>0.05 all. Compared with before treatment,after treatment,there were significant reductions in thickness of carotid plaques [left:(0.104±0.103)mm vs.(0.044±0.031)mm,right:(0.111±0.093)mm vs.(0.055±0.046)mm]in elec- troacupuncture + massage group,and they were significantly thinner than those of routine group [left:(0.190± 0.175)mm,right:(0.223±0.183)mm],P<0.01 all;significant reductions in carotid IMT [left:(0.096±0.021) mm vs.(0.086±0.019)mm,right:(0.091±0.019)mm vs.(0.087±0.018)]in electroacupuncture + massage group,and they were significantly thinner than those of routine group [left:(0.105±0.016)mm,right:(0.103± 0.020)mm],P<0.05 or <0.01.Conclusion:Electroacupuncture combined point massage can effectively reduce thickness of carotid plaques and carotid intima-media thickness.

ABSTRACT

OBJECTIVE:To compare clinical efficacy,safety and economy of urokinase and alteplase each combined with Enoxaparin sodium in the treatment of acute ST segment elevation myocardial infarction (STEMI). METHODS:80 STEMI pa-tients were randomly divided into control group and observation group,with 40 cases in each group. Both groups received Aspirin enteric-coated tablet for antiplatelet aggregation,and Clopidogrel hydrogen sulfate tablet for anticoagulation before thrombolysis. Control group were given urokinase 1 500 000 U added into 0.9% Sodium chloride injection 100 ml,ivgtt,within 30 min;given Enoxaparin sodium injection 7 500 U intramuscularly 12 h after thrombolysis,for 3-5 d. Observation group was given Enoxaparin sodium 60 U/kg,ivgtt,1 mg/ml Alteplase for injection 8 ml with intravenous push,other 42 ml ivgtt within 90 min;continued to receive Enoxaparin sodium with 12 U/(kg·h)micro-pump for 48 h,followed by Low molecular weight heparin calcium injection 5 000 U intramuscularly,bid,for consecutive 5 d. Clinical efficacy of 2 groups were observed,and thrombolytic recanalization situa-tion were observed 30,60,90 and 120 min after thrombolysis. ECG,cost-effectiveness and ADR were also observed. RESULTS:The effective rate of observation group(92.50%)was significantly higher than that of control group(85.00%). The rates of throm-bolytic recanalization in observation group 60,90 and 120 min after thrombolysis were significantly higher than in control group, with statistical significance(P0.05). The incidence of ventricular aneu-rysm,pericardial effusion,heart failure or cardiac shock,angina pectoris after infarction,severe arrhythmia,death and other as-pects in observation group were significantly lower than in control group,with statistical significance(P<0.05). CONCLUSIONS:Alteplase and enoxaparin thrombolysis therapy is better than urokinase and enoxaparin for STMEI in respects of clinical efficacy, and thrombolytic recanalization with less ADR and better safety;urokinase is cheaper and better than alteplase in cost-effectiveness ratio. Both of them can be used after careful consideration.

ABSTRACT

OBJECTIVE:To investigate the effects of Shenmai injection combined with sodium ferulate on infarct size and relat-ed indicators of patients with acute myocardial infarction. METHODS:110 patients with acute myocardial infarction were randomly divided into control group(55 patients) and observation group(55 patients). Patients in the control group received oxygen inhala-tion,coronary expansion by nitrates,water and electrolyte regulation and acid-base balance regulation,thrombolytic fibrinolytic agent utilization,sedation and analgesia by analgesics and other conventional treatments. Meanwhile,50 ml Shenmai injection was given to the control group,dissolved in 200 ml 5% Glucose injection,intravenously,twice a day;observation group was addition-ally given 0.2 g Sodium ferulate for injection,dissolved in 200 ml 5% Glucose injection,intravenously,twice a day. The treat-ment course for both groups was 14 d. Clinical efficacy in 2 groups was observed,myocardial infarction size before and after treat-ment was recorded,hemorheology indexes(whole blood viscosity,fibrinogen and erythrocyte aggregation index),catdiac enzymes index homocysteine(Hcy),and growth arrest-specific gene product 6(Gas6)before and after treatment together with the incidence of adverse reactions were detected. RESULTS:The total effective rate in observation group was significantly higher than it in con-trol group. The difference was statistically significant(P0.05). After treatment,the myocardial infarct size in 2 groups was reduced,hole blood viscosity,fibrinogen and erythro-cyte aggregation indexes were reduced,and these in observation group were lower than in control group;in the 8 day during the treatment,serum level of Hcy in observation group was reduced;in the 15 day,blood level of Hcy and Gas6 in observation groups was reduced,serum level of Hcy in control group was reduced,and these in observation group was lower than in control group. The differences were statistically (P<0.05). And there were no obvious adverse reactions during treatment. CONCLU-SIONS:Based on conventional treatment,Shenmai injection combined with sodium ferulate shows significant efficacy in the treat-ment of acute myocardial infarction,and can reduce blood level of Hcy and Gas6,improve hemorheology indexes and narrow the myocardial infarct size.

ABSTRACT

Objective:To prepare monoclonal antibody ( mAb ) against human testis-specific conserved gene ( hTSC29 ) peptides and characterize its immunological and biological features.Methods:According to bioinformatics analysis and prediction of the antigenicity, surface property, hydrophilicity and flexibility of hTSC29, a 18-amino acid residue partial peptide of hTSC29 was synthesized,then immunized the BALB/c mice for preparing antiserum.The mAb against hTSC29 was produced using the routine hybridoma technique.The properties of the mAb against hTSC29 were identified by ELISA, Western blot and immunohistochemistry staining.Results:After cell fusion and subcloning, one hybridoma cell lines secreting specific mAb against hTSC29 protein were obtaind.The Ig subclass of the mAb was IgG2b(κ).ELISA detection showed that the titer of mAbs in cultured was 1∶104.Western blot analysis proved that the mAb could specifically recognize Mr 60 000 protein in human testis total protein.The hTSC29 protein main located at circumference of spermatocyte and spermatid in human testis tissue by immunohistochemistry staining and immunofluorescence assay.Conclusion:One hybridoma cell lines which can secrete specific mAb against hTSC29 protein with high titers and specificity have been established successfully.The mAb will provide efficient tools for functional studies of hTSC29 expressed in spermatogenesis.

ABSTRACT

Objective To study the prognostic factors of gastrointestinal stromal tumors (GIST).Methods A total of 144 GIST patients treated at the First Affiliated Hospital of Guangxi Medical University between January 1996 and December 2011 were analyzed restrospectively.Kaplan-Meier,log-rank test and Cox regression model were used.Results The overall 5-year survival was 59.6%.Log-rank univariate survival analysis showed that the primary location,tumor size,metastasis,operational method,type of tumor cells,invasion of the surrounding organs and tissues,invasion of the smooth muscle,cellularity of the tumor,mitotic counts of the tumor cells,Ki-67 labeling index,P53 expression and targeting therapy were significant prognostic factors (P < 0.05).Cox regression model showed that tumor size,metastasis,operational method,surrounding organs and tissues invasion,cellularity of the tumor,mitotic counts of the tumor cells,P53 and targeting therapy were essential to improve the survival.Conclusions The study suggests that early diagnosis and comprehensive treatment consisting of operation and targeting therapy can significantly improve the survival of GIST.

Objective To test psychometric properties of the PTSD checklist-civilian version(PCL-C) among junior school students from earthquake-hit region.Methods PC L-C Chinese Version, Rosenberg self-esteem scale and SCL-90 were utilized to assess 560 junior school students from earthquake-hit region.Results Firstly,the internal consistency of the total scale and three subscales were qualified, the coefficient alphas were 0.890,0.734,0.789 and 0.722, respectively; 307 participants, from the first samples, engaged in the second test two months after the first assessment.The test-retest stability was re-experiencing 0.40(P < 0.01 ) ,avoidance 0.45 (P< 0.01 ), hyper-arousal 0.448 (P < 0.01 ), total scale 0.535 (P < 0.01 ), respectively.Secondly, the results of criteria validity showed PCL-C and its three subscales had negative correlation with self-esteem (r =-0.192 ~-0.299, P < 0.01 ), and had positive-related with SCL-90 and its subscales of depression, anxiety, and psychotic (r=0.563 ~0.775, P<0.01 ).Finally,the results of empirical validity revealed that based on the score on PCL-Ccould differentiate those people who have significant different scores on SCL-90.Conclusions The PCL-C hadgood reliabilities and validities,which could be used to assess the severity of PTSD.

ABSTRACT

Objective To investigate the protective effects of α-1 antitrypsin on human islets injured by protease released from pancreas exocrine cells. Methods ( 1 ) in vivo experiment. Parts of the cadaveric pancreas was digested with collagenase, islets were selected artificially, and pancreatic exocrine cells were collected. 8-9 weeks olds male BALB/c-Nu nude mice were induced into diabetic mice with STZ (240 mg/kg body weight, i. p) and randomly divided into two groups: the control group (n = 6), 250 islets were transplanted into left kidney subcapsule of diabetic nude mice; cotransplant group (n = 7), 250 islets and the equal volume of pancreatic exocrine cells were transplanted into different regions of left kidney subcapsule. Blood glucose level was monitored. Nephrectomies were performed after 28 days. The expression of anti-amylase antibodies in subcapsule was detected by using immunohistochemical staining. (2) Islets culture: Three groups were randomly set up. Group 1: purified islet group, 250 islets were incubated into a 6-well culture plate; Group 2: non-purified islet group, 250 purified islets and equal volume of exocrine cells were incubated; Group 3: nonpurified islet + Al AT group, 250 purified islets and equal volume of exocrine cells were incubated with α-1 antitrypsin added (0. 5 mg/ml). After 48 h, insulin content of islets in each well and trypsin concentration in the supernatant of each well were measured. Results 10000 islets were collected.After islets transplantation, the blood glucose levels in control and co-transplant groups were normal,but a delayed islet function in reversing diabetes was in the co-transplant group, and ehe mice in both groups became hyperglycemic after nephrectomy. A large number of anti-amylase antibody-positive cells were found in renal subcapsule in the co-transplant group while little seen in the control group.Insulin levels in the non-purified islet group were decreased as compared with purified islet group,those in the non-purified islet group + A1AT group were higher than in the non-purified islet group,but lower than in the purified islet groups. Trypsin concentration in the non-purified islet group was increased as compared with purified group, that in the non-purified islet group + A1AT group was lower than the non-purified islet group, but higher than in the purified islets group (all P<0. 01).Conclusion Protease released from acinar cells during pancreatic digestion has detrimental effect on islet function after transplantation. Co-cultivation of islets and pancreatic exocrine cells with A1AT added can prevent islet cell damage caused by trypsin.

ABSTRACT

To investigate the effect of NVP-DPP728, a DPP-Ⅳ inhibitor on new-onset diabetes and the autoimmune response in non-obese diabetic ( NOD ) mice. Diabetes could be reversed in 75% of NVP-DPP728 treated 20 NOD mice. In these 15 mice with remission, insulitis scores were significantly lower than those of the control group. The percentage of Tregs was increased in the thymus and celiac lymph nodes, plasma TGF-β1 and GLP-1 were also significantly increased ( P<0. 01 ). NVP-DPP728 treatment may reverse new-onset diabetes in NOD mice by reducing insulitis and increasing Tregs.

ABSTRACT

Objective To investigate the condition of inflammation and prethrombotic state (PTS) of the aged patients with coronary heart disease(CHD) complicated with esstential hypertension(EH) and type 2 diabetes mellitns(T2DM). Methods The levds of fasting plasma hs-CRP,D-D and VWF were all measured in 86 CHD patients with EH,84 patients with T2DM,80 patients with EH and T2DM,91 CHD patients and 79 normal cases. Results Compared with normal group,diseases groups had significantly higher hs-CRP,D-D,and VWF,but compared with CHD with EH and T2DM group,other diseases groups had lower hs-CRP,D-D and VWF(P <0.05) ;The concentration of serum hs-CRP was positively correlated with D-D and VWF in CHD group (P < 0. 05). Conclusion The inflammation and hypercoagulable state is more significant in CHD patients with EH and T2DM,thns the levels of plasma hs-CRP,D-D and VWF play an important role in the treatment.

ABSTRACT

PEG (Mn = 1000)-segmented polyurethanes, with hard segment percentage of 40%, 50% and 55% and named PU-H40, PU-H50 and PU-H55 respectively, were synthesized by bulk polymerization. The structure of PU was characterized by FTIR, DSC, and GPC. Mechanical properties, water contact angles and water vapor transmit rate(WVTR) were also tested. FTIR and DSC showed that the degree of microphase separation increased with the hard content. Mechanical test showed the tensile strength of PU-H50 to be 25 MPa, the highest tensile strength of the PU series. By the use of PEG as soft segment, the surface hydrophilicity of the materials increased dramatically. Owing to its high degree of microphase separation and the mobility of soft segment, the water contact angle of PU-H55 attained to 33 degrees. The WVTRs of PU-H40, PU-H50 and PU-H55 were 789 g/m2/24h, 705 g/m2/24h and 623g/m2/24h respectively. These data suggest that the materials are suitable for fabricating such biomedical articles as surgical gloves, wound dressing and medical protective coating.

ABSTRACT

It has been well known that fluorinated polyurethanes exhibit unique low surface energy, biocompatibility, biostability and nonsticking behavior. Consequently, these polymers have attracted considerable interest. In this study, the effect of various concentrations of fluorinated polyurethanes in the polyurethanes on the surface structures of the blends and their hemocompatibility were investigated by XPS, AFM, contact angle and platelet adhesion. It was found that the high concentration fluorine on the outer surfaces of the blends obtained with the low concentration of fluorinated polyurethanes (F: 0.342 wt%) in the blends was the same as that of the fluorinated poly(ether urethane)s, and all of the blends and the fluorinated poly(ether urethane)s had good hemocompatibility, compared with poly(ether urethane)s. The polymer blends and fluorinated poly(ether urethane)s suppressed platelet adhesion due to their high hydrophobicity and low surface tension. The XPS, AMF and contact angle results indicated that the high hydrophobicity of outer surface of the polyurethane blends is independent of the fluorinated polyurethanes content in the polymer blends but related to the concentration of the CF3 groups because the lower critical surface tensions and higher contact angle of many fluorinated surfaces reflect the concentration of CF3 groups.

ABSTRACT

The use of biodegradable polyurethane as medical appliance has rapidly become a fascinating new field of research in biomedical science. Tremendous progress has been made in this area during the last years. Applications of these materials in the field of organic repairing, tissue engineering and drug-controlled delivery system were reviewed. These new type materials possess excellent properties, such as good biocompatibility and mechanical strength, facile formation and inexpensive, so it can be expected to find wide application in the medical treatment in future.

ABSTRACT

In this article, the blood compatibility of polyurethanes (PUs) made by ourselves for catheters is studied by hemolysis test, platelets adhesion test, kinetic thrombus time test and dynamic clot formation. The results showed that these PUs all have excellent blood compatibility. Among these PUs, H50-100 and H60-100 have best blood compatibility. Additionally, the relationship between the structure and blood compatibility is discussed.

ABSTRACT

Polyurethanes are popularly used in cardiovascular and other biomedical fields due to their good biocompatibility as well as mechanical properties. But they are subject to biodegradation in vivo for a long time, and cause inflammation, so improving the biocompatibility of medical polyurethanes is an important subject of biomaterials. Recent researches have focused on biological modelling of biomaterials for improving the biocompatibility of polyurethanes. This paper reviews two main methods for improving biocompatibility of polyurethanes-endothelial cells seeding and mimic biomembrane (phospholipid surface), and summarizes the main procedures and questions of these two methods.
